The Alcian Yellow Toluidine Blue (AYTB) Kit is a specialized histochemical staining system designed to demonstrate acidic mucopolysaccharides and glycosaminoglycans in tissue sections. This dual-stain approach combines Alcian Yellow for sulfated mucins and Toluidine Blue for acidic mucopolysaccharides, providing excellent differentiation for connective tissue studies, cartilage analysis, and mucin-secreting tumors. The kit produces distinct color reactions that enable clear visualization of mucin types in gastrointestinal, respiratory, and connective tissue samples.

Staining Characteristics

AYTB Staining Results

Sulfated Mucins

Stain yellow with Alcian Yellow. Strong staining indicates sulfated glycosaminoglycans found in cartilage and mast cells.

Carboxylated Mucins

Stain blue with Toluidine Blue. Indicates sialomucins and hyaluronic acid in connective tissue and epithelial cells.

Metachromasia

Toluidine Blue demonstrates metachromatic properties with heparin and cartilage matrix, staining purple-red.
