Alcian Blue Kit

Acid Mucopolysaccharide Stain โ€” Mucin Detection

Special Stain Kit

Alcian Blue Kit

CE-IVD Certified Histochemical Grade โ€” Supplied by Delonix Enterprise

Alcian Blue is a widely used histochemical stain that specifically demonstrates acid mucopolysaccharides and mucins in tissue sections. The copper-phthalocyanin dye forms salt bonds with acidic tissue groups, staining mucopolysaccharides an intense blue color. This kit is essential for identifying epithelial mucins in gastrointestinal tract samples, respiratory mucosa, and connective tissue matrices. Available in pH 2.5 for weakly acidic mucins and pH 1.0 for strongly acidic (sulfated) mucins formulations.

Stain Type
Mucopolysaccharide / Mucin
Target
Acid mucopolysaccharides & mucins
pH Options
pH 1.0 (strongly acidic) | pH 2.5 (weakly acidic)
Mechanism
Copper-phthalocyanin salt bond formation
Results
Acid mucopolysaccharides: Blue | Nuclei: Pink/Red
Kit Includes
Alcian Blue solution + Nuclear counterstain
๐Ÿ’ก Differentiate between sulfated and non-sulfated mucins using pH variations. At pH 1.0, only sulfated mucins stain; at pH 2.5, both carboxylated and sulfated mucins stain blue.
Staining Characteristics

Alcian Blue Results

1

pH 2.5 Staining

Stains both carboxylated (sialomucins) and sulfated mucopolysaccharides blue. Used for general mucin screening.

2

pH 1.0 Staining

Specific for strongly acidic sulfated mucins only. Differentiates sulfated from non-sulfated mucopolysaccharides.

3

Nuclear Counterstain

Included nuclear fast red or neutral red stains nuclei pink-red for contrast against blue mucin deposits.

4

Clinical Applications

GI biopsies, bronchial mucosa, cartilage studies, and identification of mucinous carcinomas.
