PAS Kit

Periodic Acid-Schiff โ€” Carbohydrate Detection

Carbohydrate Stain Kit

Periodic Acid-Schiff (PAS) Kit

Research & Diagnostic Grade โ€” Supplied by Delonix Enterprise

The Periodic Acid-Schiff (PAS) reaction is a fundamental histochemical technique for demonstrating carbohydrate-rich macromolecules in tissue sections. The kit stains glycogen, mucins, glycoproteins, and proteoglycans a brilliant magenta color. Essential for diagnosing glycogen storage diseases, identifying fungal organisms, highlighting basement membranes, and detecting mucin-producing tumors. The PAS technique can be combined with diastase digestion to specifically confirm glycogen presence. This versatile stain is indispensable in renal pathology, liver biopsies, gastrointestinal studies, and dermatopathology.

Stain Type
Carbohydrate / Glycoprotein
Target
Glycogen, mucins, glycoproteins, fungi, basement membranes
Mechanism
Periodic acid oxidation โ†’ Schiff reagent reaction
Applications
Liver, kidney, GI tract, fungi detection, glycogen storage
Results
PAS-positive material: Magenta | Nuclei: Blue (with hematoxylin)
Kit Includes
Periodic acid solution, Schiff reagent, hematoxylin counterstain
๐Ÿ’ก Use PAS with diastase digestion to differentiate glycogen (diastase-sensitive) from other PAS-positive substances (diastase-resistant). Essential for diagnosing glycogen storage diseases like von Gierke disease.
Staining Characteristics

PAS Staining Results

1

Glycogen

Stains bright magenta. Digestible by diastase - confirm by comparing digested vs undigested sections.

2

Mucins

Epithelial mucins stain magenta. Combine with Alcian Blue to differentiate neutral vs acidic mucins.

3

Fungi

Fungal cell walls contain polysaccharides that stain magenta. Essential for detecting Candida, Aspergillus.

4

Basement Membranes

GBM and tubular basement membranes stain magenta. Critical for evaluating kidney biopsies.
